If you're a new blogger, you might want to go direct to WP 2.5.

If you've a legacy blog (ie WP 2.3 or earlier), you might want to set up a test environment (ie a /test/ subdirectory or prefably a domain you're not using), move the MySQL files over, together with the plugins and templates you've been using and do the migration to 2.5. disable the ping server notification, so you don't have issues with dupe content and see if your legacy plugins work.
If necessary, upgrade to the latest version of your plugins. (In some cases, these plugins are still not WP 2.5 compatible).

There's been coverage that Technorati is not indexing blogs priori to WP 2.5, although I found that this hasn't been the case for me, because I'm using WP 2.3.x and still getting indexed.
The most important point to remember in all of this is to backup your MySQL database which contains your blog posts and pages. If that's shot, there goes your blog.
