I believe a suppression list is a list of people who have opted out of a mailing list. You are supposed to compare future mailing lists against the suppression list to make sure you are not sending unsolicited emails to people who have opted out.
Sending emails to people on a suppression list would count as spam and would be illegal.
I dont know the ins and outs of it but that seems to be the gist.
