A few approaches:
Use a captcha system - make the user answer one of those silly puzzles like "type the random text that's slightly difficult to read." Users don't really like them, but typically understand.
Make a hidden comment form that does nothing, and place it higher up in the html than your real comment form. Most spam bots will will just fill out the first form they encounter and go happily on their way.
Nofollow any comment link for a few days. If it survives moderation, turn it into a normal link. That will make you a much less attractive target, but it'll take the longest to see any effect.
