While many people do read from left-to-right, there are languages like Arabic and Hebrew that are the opposite. However, for most western languages, the upper left is a natural starting point. That said, it really is site specific as Nick mentioned. If you study design at all, you'll learn how to draw the eye to your desired location. You can use a lot of subtle techniques like contrast in size or color, whitespace, images, font, or motion (be careful with motion). Rather than just blindly following someone's advice, the best thing you can do is test on your site to see what works with your design. A/B and multivariate testing will help you optimize the conversion rate of your site, no matter if you are just serving ads or trying to sell products.
