Can anyone please tell me that which one is the most important and beneficial on page optimization activity nowadays? and how it is helpful according to Google?

Firstly, check what your competitors are doing, research your keywords and see what OnPage SEO they've implemented. SEO is basically doing what your competitors are doing, just doing that little bit better...
Ok, on your page you should have a TITLE like this:
Company Name | Service 1, Service 2
You should have a matching H1 Tag like:
Service 1, Service 2 from Company name
Then you need a well written DESCRIPTION. All of these three elements should contain your keywords/phrases.
Then you should have your keywords in the first paragraph of your text. Run your page through a Keyword Density Checker, make sure your keywords are the biggest.
Other points:
Code your page well, make it fast loading
Utilise semantic web design
Have text based navigation
Have a Google sitemap
Install Google's webmaster tools
Make good use of image ALT tags.
In all of the above, DO NOT just stuff lots of keywords in there. Focus on 3 and synonyms of them. Don't go crazy...

Following are 3 imp points for onpage activity -
1- look at your competitors’ websites to see what keywords they are targeting
2 -Review their meta keywords, titles, and content on their websites to identify additional keywords to add to your list.
3- look for keywords is forums, blogs, industry sites, and Q&A sites like Yahoo Answers

If you have images on your web page then use your keyword in image url and alt tags. You will see a boost in your ranking in SERP.

There is no just one single most important on page activity. But there is a set of important on page activities. This set includes
1) Proper keyword research
2) Competitive analysis.
3) Keywords in Title & Meta Tags
4) Keywords in Content with reasonable keyword density
5) For Images, Place Keyword-rich Alt & Title Tags

I agree with all of the above - "sprinkle" keywords in a "natural" fashion through title tag, h1-h6 tags, alt text, content, page urls, image names etc.
Some say the description tag is not important for seo but needs to be written to encourage users to click on your site when it appears in the serps; putting keywords in the Description whilst still making it attractive to read is probably a safe bet.
I have also found that it helped my site when I changed a repeating sidebar to a left-hand column that was unique for each page (unique content, optimised for each page) and also I made the navigation links to less important inner pages nofollow so that there was more link juice available to some of the inner pages I was targetting.

In my point of view interlinking of webpages is also very important
interlinking through the content of pages is more effective as compare to side menu linking.

in my opinion there is no such thing as very important when it comes to seo, for me all of the techniques and strategies are important so you have to apply those and just eliminate which are not effective to your site.

You should have the right keyword density in your article content on a single page or at least be on the rule of having a single target keyword in once in every 100 words. Place your target keyword in the title and on the first sentence. This is what search engines love.
If you are targeting your site to have adsense ads, you should place lots of those keywords in the meta tags of the site for Google to supply the best targeted ads.

Keywords are the basi of SEO, select the less Competitive and good traffic, and then edit a good title! Like xxx | xxxxxsx, so this can attract google and other SE!

The canonical thing is hard to understand, even after reading up on it. Should you add it when you have pages that just share 50% duplicate content with each other? or 25% ?
It seems like lots of work, but might be effective.
