Before starting SEO, you need to research your keywords with a tool such as Google analytics keyword tool then - preferably - optimise domain name and also optimise the title tag, h1-h6 tags, alt text, content, folder names, file names, image names, navigation etc. Don't overdo this or you could get a penalty for spamming. Use one of the keyword density tools (Google this) to check you've included your keywords but not too much.
Some say that you should also put keywords into the description and keyword tags, other say this is no longer relevant.
See a beginner's guide to onpage seo for a discussion of how to optimise each part and also sample code where relevant.
once you've got your onpage seo in palce,l start building backlinks with your keywords as your anchor text - but make sure you mix up the keywords otherwise your links could get ignored. The aim is to make the backlink campaign seem "natural" with a mix of links from different types of sources (artidcles, blogs, directories, forums etc), different page ranks, a few nofollow links and a few links with non-keyword anchor text such as "my site".

Well, there are a couple of things that I look at.
- Keyword research
- information architecture (Page and URL structure/ layouts, etc)
- Title/headings/ALT tags/anchor text, etc
- Keyword frequency
Read the following. The article is better than I can explain it. Great article

I have to agree with a lot of what was already said here. The domain name should have the main word that you wish to get ranked for in it. The main word or key word should be in the article, blog etc. I would make the keyword bold a few times through out the content but don't over do it. Get decent back links and it will rank. Not as hard as it seems but just a lot to remember when starting out when it pertains to seo.
